Project: I am working on the site-specific synthesis of structural analogues of the N-(Deoxyguanosin-8-yl) aminofluorene adduct containing a functionalized linker that would allow their site-specific fluorescent labeling by coupling with commercially function-specific activated fluorescent dyes after their incorporation into DNA by solid phase DNA synthesis. Such fluorescent and proficient NER substrates will then allow the study of this DNA repair machinery using fluorescent techniques. Those adducts will be used to monitor the interaction of the individual NER proteins by measuring the Föster Resonance Energy Transfer of these lesions within GFP fused NER proteins.
